Agilent 5200 Fragment Analyzer – Fast, accurate, high-throughput DNA/RNA analysis
The Agilent 5200 Fragment Analyzer is a fully automated capillary electrophoresis (CE) system for the separation and quantification of double-stranded nucleic acids, whether DNA or RNA. Its technology is based on the use of an electrophoresis beam applied to a network of fused silica capillaries, each filled with a specific conductive matrix. This matrix is optimised to sort molecules according to their size, providing accurate visualisation of the distribution of fragments present in the samples. Separation is achieved quickly by applying an electric field, with the smallest fragments migrating faster than the larger ones.
The system incorporates a fluorescence detection module based on a high-intensity LED light source focused on the capillary detection window. DNA or RNA fragments labelled with a fluorescent intercalating dye are detected by a high-sensitivity CCD camera, generating digital electropherograms that can be used in real time. The accuracy of the measurement provides clear and reproducible migration profiles, which are essential for applications in genomics, transcriptomics or sample preparation quality control.
The 5200 Fragment Analyzer is designed for flexibility and efficiency. It can be configured with a network of 12, 48 or 96 capillaries to suit the volume of samples to be analysed. The 96-well plates are easy to load thanks to an ergonomic drawer system, each drawer dedicated to a specific function (buffer, marker, samples, waste). This architecture simplifies handling and reduces the risk of positioning errors, while maintaining a high level of safety thanks to interlocking systems that prevent inappropriate opening during high-voltage operations.
The analysis process is fully automated by the associated Fragment Analyzer software, which is compatible with the ProSize tool for data interpretation. Users can configure and launch their analysis sequences, monitor separations in real time, and utilise the results via automated reports. The graphical interface facilitates method tracking, parameter control and data integration, ensuring quick start-up even for non-specialised operators.
The integrated high-pressure pumping module ensures automatic preparation and conditioning of the capillaries between each series of analyses. This step, which includes rinsing, filling with separation gel and removing residues, extends the life of the capillaries and guarantees consistent performance. The system also offers a choice between electrokinetic or vacuum aspiration injection, which is particularly useful for samples rich in salts or containing potential inhibitors.
Compact and robust, the 5200 Fragment Analyzer is ideal for research laboratories, sequencing platforms or quality control units.
