The system features an intuitive method wizard that streamlines protocol programming for a wide range of chromatographic techniques, including ion exchange (anion or cation), hydrophobic interaction, affinity chromatography, gel filtration, and chromatofocusing. With an extensive library of pre-defined columns, the system automatically optimizes parameters for each selected column, ensuring overpressure protection and instant operation without prior expertise. The built-in scouting function allows rapid method development and optimization by varying key parameters such as flow rates, sample volumes, or gradient profiles.
ÄKTAFPLC delivers consistent reproducibility and high analytical sensitivity. Its dual-piston P-920 pump ensures pulse-free flow with precise pressure control up to 5 MPa, enabling reliable separations even with viscous solvents. The integrated UPC-900 monitor performs in-line detection of UV absorbance (at 254, 280, and optionally 214 nm for enhanced sensitivity), conductivity, and, optionally, pH, with automatic temperature compensation.
Sample injection can be performed manually or automatically via the A-900 autosampler, which accommodates up to 160 micro-samples. The system also supports Superloop injection loops up to 150 ml, offering flexible volume handling. Fraction collection is managed by the Frac-950 collector, compatible with all tube and plate formats, enabling automatic peak-based fractionation to minimize dilution and cross-contamination.
Highly modular, the ÄKTAFPLC allows seamless integration of additional components, such as air sensors, column or buffer selection valves, and automation accessories, all centrally controlled through UNICORN software.
