Description

Enhance Your Analytical Capabilities with the CESI 8000 Plus: Multi-MS Connectivity and Multi-Detection Capability for Comprehensive Biomolecular Analysis

The Sciex CESI 8000 mass spectrometer is a cutting-edge instrument designed to offer unparalleled capabilities in the analysis of charged and polar compounds in various analytical laboratories. With the CESI 8000 Plus High Performance Separations-ESI Module, it becomes an indispensable tool for researchers seeking accurate and detailed insights into complex biological samples.

This advanced accessory excels at consuming minute samples with extraordinary sensitivity, making it ideal for the analysis of charged and polar metabolites, peptides, and more. Its high-resolution separation capabilities extend to proteins and protein-complexes, allowing researchers to delve deeper into the composition of biological samples.

One of the standout features of the CESI 8000 Plus is its ability to eliminate ion suppression bias, providing researchers with a clearer view of their samples and revealing details that may have been previously obscured. Despite its sophisticated capabilities, the instrument remains robust and easy-to-use, ensuring accessibility for researchers of all levels of expertise.

The CESI-MS technology is particularly valuable for analyzing precious samples, including subcellular fractions, microdialysates, CSF, murine samples, CTCs, needle biopsies, FFPE archives, and highly toxic samples such as ADCs and venoms. Its unique multi-segment injection system significantly increases throughput by up to 10X, allowing researchers to analyze more samples in less time.

Furthermore, the CESI 8000 Plus offers extraordinary sensitivity and high resolution, enabling maximum information retrieval with minimal sample consumption, typically requiring only around 50 nL per injection. For example, using just 100 ng of yeast mitochondrial extract, CESI-MS has shown to significantly increase peptide identification, resulting in a substantial increase in the number and variety of proteins identified.

Measurements of highly charged analytes present significant challenges for existing technologies. However, CESI emerges as the method of choice for achieving high-resolution separation of polar metabolites, peptides, and proteins rich in specific post-translational modifications, such as glycosylation, citrullination, methylation, and phosphorylation. Developed in collaboration with mass spectrometry researchers across various applications, the CESI 8000 Plus was designed to expand the detection range and enhance sensitivity. To accomplish this, ultra-low flow separation was integrated with mass spectrometry using proprietary CESI-MS technology.

The CESI 8000 Plus incorporates a sprayer within the protective housing of the OptiMS cartridge, combining intrinsically low-flow CE separation with electrospray ionization (ESI) in a single device. The CESI OptiMS capillary inlet and outlet maintain the same inner diameter, ensuring consistency and efficiency in the separation process. Electrical contact for both CE separation and ESI generation is achieved through the ESI metal needle filled with conductive fluid.

Ultra-low flow at the porous capillary tip instantly generates a fine ESI spray when a low voltage of approximately 1.25 KV is applied. This feature not only ensures precise and efficient ionization but also minimizes the risk of oxidation artifacts due to the low voltage employed in the CESI-MS process. Overall, the integration of ultra-low flow separation and electrospray ionization within the CESI 8000 Plus represents a significant advancement in mass spectrometry technology, offering researchers enhanced sensitivity, improved detection capabilities, and greater flexibility in analyzing complex biological samples.

The CESI 8000 Plus offers multi-MS connectivity and multi-detection capability, enhancing its versatility and applicability in various analytical workflows. In CESI-MS mode, the instrument can seamlessly integrate with industry-leading SCIEX mass spectrometers, providing researchers with access to advanced analytical capabilities and high-performance mass spectrometry technology. This integration enables comprehensive analysis of complex biological samples, including polar metabolites, peptides, and proteins with specific post-translational modifications.

Furthermore, the CESI 8000 Plus can operate in stand-alone CE mode, offering a range of detection options such as UV/VIS, PDA (photodiode array), or LIF (laser-induced fluorescence). This flexibility allows researchers to tailor their analytical approach based on the specific requirements of their experiments. For instance, the instrument can be utilized for the analysis of biologics using techniques such as CE-SDS (capillary electrophoresis-sodium dodecyl sulfate), cIEF (capillary isoelectric focusing), and glycan chemistries.

By offering multi-MS connectivity and multi-detection capability, the CESI 8000 Plus empowers researchers to explore diverse analytical techniques and methodologies, enabling comprehensive characterization of complex biomolecular samples. Whether integrated with mass spectrometry for high-resolution analysis or utilized with various detection options in stand-alone mode, the instrument provides researchers with the flexibility and performance required to address a wide range of research questions and applications in the fields of proteomics, metabolomics, and biopharmaceutical analysis.
