TLA-55 – Fixed angle rotor for benchtop ultracentrifuges
The Beckman Coulter TLA-55 rotor is a state-of-the-art centrifugation solution designed for demanding applications in research and biotechnology laboratories. Compatible with a wide range of Beckman Coulter benchtop ultracentrifuges (Optima MAX-XP, MAX, MAX-TL, TLX, TL-100, etc.), this fixed-angle rotor enables efficient and rapid separation of biological, chemical or pharmacological components.
Madeof anodised aluminium for excellent corrosion resistance, the TLA-55 can accommodate up to twelve 1.5 mL microcentrifuge tubes at a fixed angle of 45°, the optimal position for fast pellet formation while minimising loss of biological activity. Each tube cavity is numbered for easy sample identification.
The rotor features a lid with a push-button locking system and two Buna-N rubber O-rings to ensure a tight seal during centrifugation cycles. A fluid retention ring located below the O-ring surface prevents accidental leaks from contaminating the centrifugation chamber.
In terms of safety, the TLA-55 incorporates magnets for speed detection and overspeed prevention, coupled with a magnetic sensor in the instrument. This mechanism ensures that the ultracentrifuge never exceeds its maximum permissible speed of 55,000 rpm, equivalent to a centrifugal field of 186,000 × g.
The rotor is optimised for CsCl density gradients, with precipitation curves specifically established according to temperature and density, ensuring gradient stability during centrifugation. The rotor's k factor, set at 66, allows for accurate calculation of centrifugation times based on the sedimentation coefficient of the target particles.
For safe use, preliminary checks are necessary, including inspection of the O-rings and pusher, as well as rigorous load balancing. If a leak is detected after a cycle, a decontamination procedure is mandatory.
In terms of maintenance, Beckman Coulter recommends cleaning at least once a week, regularly applying silicone vacuum grease to the seals and lubricating the threads with Spinkote. The components can be sterilised in an autoclave at 121°C for one hour.
